ultrasound assisted extraction of curcumin

Product Detail

Introduction

Curcumin is the main lipophilic polyphenolic active ingredient in turmeric (Curcuma longa), which has strong antioxidant, anti-inflammatory, and potential anti-cancer effects. However, its poor water solubility, low oral bioavailability, and easy photodegradation and pyrolysis limit its direct application. Therefore, efficient, gentle, and green extraction techniques are crucial. Ultrasonic assisted extraction (UAE), with its cavitation effect, can significantly damage plant cell walls and improve mass transfer efficiency, and has become one of the mainstream green technologies for extracting curcumin.

Advantage
UAE achieves a good balance between efficiency, cost, and sustainability. Compared with traditional extraction methods (time-consuming and low yield), Soxhlet extraction (high-temperature and long-term, easily degradable), and microwave-assisted extraction (MAE, local overheating risk), UAE can shorten the extraction time to minutes while maintaining the structural integrity of curcumin, and increase the yield by 2-5 times. In addition, UAE and enzymatic methods NADES、 Combined strategies such as spray drying (such as "UAE+enzymolysis pretreatment" or "UAE extraction+spray co embedding") are becoming a new paradigm to improve the stability and bioavailability of curcumin

1)The efficiency is 3-5 times higher than the traditional Soxhlet extraction method, and the time is shortened by 70%;
2)Low temperature operation preserves the activity of thermosensitive components

Ultrasonic assisted extraction process of curcumin

1) Raw material pretreatment
Drying and crushing: After washing and drying, the turmeric rhizome is crushed to 40-60 mesh (particle size 0.3-0.45mm) to increase the specific surface area and improve extraction efficiency.
Enzyme assisted treatment (optional): Some processes use cellulase or pectinase pretreatment to disrupt cell wall structure. Optimization conditions: pH 4.5, temperature 55 ℃, enzyme dosage 21mg/g, enzymatic hydrolysis for 42 minutes.
2) Solvent selection and ratio
Common solvents: ethanol (concentration 60% -80%), petroleum ether, ether, or mixed solvents (such as Tween-80: ethanol: water=2.5:67.5:30).
Liquid to solid ratio: 1:20~1:30 (g/mL), excessive ratio may reduce the extraction rate.
Core steps of ultrasound extraction
Parameter optimization:
Power: 200-300W (too high may cause thermal degradation);
Temperature: 40-50 ℃ (exceeding 60 ℃ can easily cause curcumin decomposition);
Time: 40-60 minutes (insufficient extraction if too short, decreasing efficiency if too long).
Dynamic extraction technology: using a continuous flow ultrasonic device with a solvent flow rate of 0.4mL/min to achieve industrial continuous production.
Post processing and purification
Solid liquid separation: The extraction solution is vacuum filtered or centrifuged to remove residue.
Concentrate and purify: After rotary evaporation concentration, curcumin with a purity of over 95% is obtained through macroporous resin adsorption, silica gel column chromatography, or crystallization purification.
Drying: spray drying or freeze drying of finished products

Application field
Medicine and Health Products
Liver protection: Curcumin liver protection tablets (such as KNC Ganyilai) reduce transaminase (ALT ↓ 40.57%) and improve fatty liver, with a repurchase rate of 98.6%.
Anti inflammatory and tissue repair:
Curcumin pretreatment of mesenchymal stem cell exosomes (cur exo) inhibits ferroptosis through the miR-16-5p/Smad3/Mb axis and alleviates renal ischemia-reperfusion injury;
Enhance lung barrier function and alleviate high-altitude acute lung injury.
Anti cancer adjuvant: Nanoparticles encapsulating curcumin enhance bioavailability and prolong circulation time in the body.
food industry
Functional additives:
Adding 0.1% -0.5% curcumin to animal feed can increase the slaughter rate of broiler chickens, reduce abdominal fat deposition, and improve pork quality;
Natural pigments replace synthetic pigments and are used in curry powder, beverages, etc.
Extension of Extraction Technology: Similar processes are applied to the extraction of natural products such as vanillin and polysaccharides.
Cosmetics and daily chemical products
Extraction of low aromatic components: Petroleum ether is used as a solvent to extract low volatile compounds, suitable for sensitive muscle anti-inflammatory formulas.
Antioxidants: eliminate free radicals and delay skin aging.
